I used to smoke. Would have paid more for insurance if I wanted to continue. I'm not poor and understand decisions have consequences.

    Hey everyone.  Im still in the hospital.  No ventilator.  No visitors.  Breathing treatments multiple times a day. Chest xrays every other day. Pulse oxygen is 89% with a nonrebreather mask running fu

  • Update  Surgery was a success. Mom has been home since this afternoon. Some pain, but good otherwise and they got the entire tumor.  Thanks all for the well wishes and prayers.

The spike (S) protein of Severe Acute Respiratory Syndrome-Coronavirus-2 (SARS-CoV-2) binds to a host cell receptor which facilitates viral entry. A polybasic motif detected at the cleavage site of the S protein has been shown to broaden the cell tropism and transmissibility of the virus. Here we examine the properties of SARS-CoV-2 variants with mutations at the S protein cleavage site that undergo inefficient proteolytic cleavage. Virus variants with S gene mutations generated smaller plaques and exhibited a more limited range of cell tropism compared to the wild-type strain. These alterations were shown to result from their inability to utilize the entry pathway involving direct fusion mediated by the host type II transmembrane serine protease, TMPRSS2. Notably, viruses with S gene mutations emerged rapidly and became the dominant SARS-CoV-2 variants in TMPRSS2-deficient cells including Vero cells. Our study demonstrated that the S protein polybasic cleavage motif is a critical factor underlying SARS-CoV-2 entry and cell tropism. As such, researchers should be alert to the possibility of de novo S gene mutations emerging in tissue-culture propagated virus strains.

Author summary

SARS-CoV-2 uses its spike (S) protein to enter target cells. Unlike other similar coronaviruses, the nascent S protein has a polybasic cleavage motif and is cleaved by the host protease. We have identified SARS-CoV-2 variants with mutations at the cleavage motif of S protein (S gene mutants) which undergo inefficient proteolytic cleavage, generate smaller plaques, and infect fewer cell lines. Notably, S gene mutants emerged rapidly through SARS-CoV-2 propagation in Vero cells. Since Vero cells are commonly used for SARS-CoV-2 propagation, it is a very real possibility that researchers have performed experiments, screened antivirals, and developed vaccines using SARS-CoV-2 S gene mutants without realizing.

Studies showing people are really infectious 2 days before and 3 days after symptomatic/testing positive. Staying isolated for the full 10 days is too long... After symptoms wane, wear a mask for 5 days just to be safe.

IMO it seems like public policy is shifting towards those who are vaccinated/boosted. If you are still unvaxxed, then it's your own fault. The world isn't going to wait any longer. The virus is endemic, might as well learn to live with it. Godspeed.

Observational studies without control groups don’t show causation, that shows CORRELATION.

Correlation isn’t evidence that proves that something works.  It’s a starting point that leads to further evaluation to see if there is CAUSATION. 

I can run a study that evaluates all 32 NFL teams that shows that teams that have their QB kneel the ball in the second half are victorious 100% of the time.  
Does this mean that there is evidence that proves that kneeling the ball in the 2nd half causes NFL teams to be win games?   If an nfl team tried employing the strategy of kneeling the ball to start the 3rd quarter, do you think that would lead to them winning the game?  Or does my study simply show correlation between kneeling the football and winning football games.
